چکیده
Human stanniocalcin-1 (STC1) is a paracrine factor associated with inflammation and carcinogenesis. The role of STC1 in the pro- anti-inflammatory functions differentiating macrophage, however, not clear. In this study, our data showed that phorbol 12-myristate 13-acetate (PMA) treatment induced human leukemia monocytic cells (ThP-1) differentiation to M0 macrophages. was accompanied by significant increase mRNA expression levels STC1, pro-inflammatory cytokine TNFα, markers, CD163 & CD206. An intermitted removal PMA reduced TNFα but had no noticeable effects on markers. correlation markers macrophages investigated, using siRNASTC1-transfected PMA-induced cells. Consistently, transcripts IL-6 were significantly reduced. Moreover, LPS/IFNγ-induced M1-polarization remarkably higher than IL-4/IL-13-induced M2-macrophages M0-macrophages. Transcriptomic analysis M1-polarized revealed an upregulation TBC1 domain family member 3 (TBC1D3G). gene regulates payload macrophage-released extracellular vesicles mediate inflammation. conditioned media from found reduce Hep3B cell motility. suggest macrophage differentiation, preferentially M1 polarization.

In macrophages, changes in intracellular calcium have been associated with activation of cellular processes that regulate cell adhesion and motility and are important for the response of macrophages to antigenic stimuli. The mammalian counterpart of the fish calcium-regulating hormone stanniocalcin-1 (STC1) is expressed in multiple organs including the thymus and spleen, and hence, we hypothesi...
متن کاملCharacterization of Stanniocalcin-1 Receptors in the Rainbow Trout
Mammalian stanniocalcin-1 (STC-1) is one of several ligands targeted to mitochondria. High affinity STC-1 receptors are present on the mitochondrial membranes of nephron cells, myocytes, and hepatocytes, to enable ligand sequestration within the matrix. However, STC-1 receptors have not been characterized in fish. Nor is it known if mitochondrial targeting occurs in fish. The aim of the study w...
متن کاملExpression and in vitro Characterization of Herpes Simplex Virus 1 (HSV-1) ORF P Protein
Herpes simplex virus 1 (HSV-1) unspliced 8.3 latency associated transcript (LAT), which located in the long repeat sequences, has been shown to contain at least 16 open reading frames (ORF: A-P). One of these ORF, ORF P, maps almost entirely antisense to HSV-1 neurovirulence gene, ICP34.5. Both ORF P and ICP34.5 are located in the long repeat and are antisense overlapping genes. Therefore, in O...
متن کاملExpression of Stanniocalcin-1 and Stanniocalcin-2 in Laryngeal Squamous Cell Carcinoma and Correlations with Clinical and Pathological Parameters
BACKGROUND Stanniocalcin-1 (STC1) and stanniocalcin-2 (STC2) are secreted glycoprotein hormones involved in various types of human malignancies. The roles of STC1 and STC2 in laryngeal squamous cell carcinoma (LSCC) remain unknown. We investigated correlations between STC1 and STC2 expression and clinicopathological or prognostic factors in LSCC. METHODS Pre-surgical peripheral blood samples ...
متن کاملStanniocalcin 1 and ovarian tumorigenesis.
BACKGROUND Stanniocalcin 1 (STC1) is a secreted glycoprotein hormone. High expression of STC1 has been associated with several cancers including ovarian cancer, but its role in the development of ovarian cancer is not clear. METHODS We used five human ovarian epithelial cancer cell lines (OVCA420, OVCA432, OVCA433, SKOV3, and HEY), immortalized human ovarian surface epithelial cells (T29 and ...
